ELISA based Biosensor to detect H1N1

Hi,

I have to come up with a mini design for one my courses and I was thinking about designing an ELISA based biosensor to detect H1N1. I am confused as to what my sample should be, should I use nasopharyngeal swab or serum?  Where (and how) do I introduce the sample to my biosensor? I would really appreciate if someone could direct me to the right track.
